1. P04.102 Stereotaxic implantation of retinal pigment epithelial cells
attached to microcarriers in advanced Parkinson disease (PD) patients: A
pilot study

Implantation of dopamine-producing retinal cells into the striatum can
improve parkinsonian symptoms, according to this report. Six patients with
advanced PD (mean disease duration 10.2 years) were implanted with human
retinal pigment epithelial cells attached to microscopic cross- linked
gelatin spheres (Spheramine) without immunosuppression. Each patient
received approximately 325000 cells, placed in 5 tracts 5 mm apart on the
side contralateral to the worst symptoms. UPDRS motor off scores fell for
each patient, with an average of 17 points reduction after 3 months.
Dyskinesia Rating scores improved 60% in the same period. One patient has
been followed for a year, and two others for 6 months, with maintenance of
improvements in motor score and dyskinesia.

2. S46.001 Intraventricular administration of GDNF in the treatment of
Parkinson's disease

Fifty patients received placebo or 25-4000 micrograms intraventricular
glial cell line-derived neurotrophic factor monthly for 8 months. No
improvements were seen in on or off UPDRS motor scores, despite clear
evidence of biological effects of treatment, including anorexia, weight
loss, and sensory symptoms in most treated patients. In 1 patient who died
of unrelated causes, there was no evidence of increased tyrosine
hydroxylase-positive fibers in the striatum.
