There is a discussion in Duvoisin's book (pp 125-127) about B6 and Sinemet.
Briefly, B6 doesn't help much in PD. In the RDA dosage range, it is
counteracted by the carbidopa component of Sinemet, so it doesn't hurt much
either. However, therapeutic or megadose (10mg-20mg/day) of B6 can
overwhelm the protection of carbidopa, and therefore should be avoided. If
such dosage is warranted for reasons other than PD, the choice should be
guided by a physician.
