Sinamet is a combination of Carbadopa and L-Dopa.(25/100 = 25mg Carbadopa and 100mg L-Dopa). At L-Dopa doses above 600mg per day the chances of dyskinetic side effects greatly increase. The current thinking is to keep the L-Dopa dose as low as possible by combining it with dopamine agonists and/or Eldepryl. Soon there will be a new class of drugs available called COMT inhibitors (Tolcapone), which will also aid in keeping the L-Dopa dose low. Jerry Starr RPh.
