Dave, I am not sure what you mean by "drug kick-in", but I take sinemet CR 25/100 on an "as needed basis". That is if the tremor is uncontrollable or I must be "on" then I take it. I sometimes take it as often as one every two hours. The med does not "kick-in", but my friends and children notice that within 15 to 30 minutes after I take it my tremor lessens or stops.
