Vitamin B6 was a factor when levodopa alone was used; however, with
Sinemet (carbidopa-levodopa) normal amounts of B6 are not considered to
interfere with absorption. Very large amounts of B6 (as in some
supplement) can interfere with Sinemet, though, and should be taken
separately from Sinemet by 2 hours.

Protein also blocks Sinemet absorption, some people are more sensitive
to protein than others, however.
